Kurzfassung

Beim iLoad®-System handelt es sich um mit Wirkstoffen wiederbeladbare Textilien (Reloadable Drug Delivery Textiles, RDDT) zur transdermalen Anwendung von kosmetischen bzw. medizinischen Wirkstoffen für den Wellness- und Gesundheitsbereich: iLoad® bedeutet intelligent beladen, abgeben, waschen und wiederbeladen.
Das iLoad®-System besteht vorab aus einem wählbaren Basisgewebe. Darauf wird eine spezielle Donorschicht aufgebracht und fest fixiert. Beim anschliessenden Beladungsvorgang wird die Donorschicht (host), d.h. das iLoad®-Textil, mit einer abgestimmten Emulsion (guest) mit den individuell gewünschten, vorzugsweise phytotherapeutischen Wirksubstanzen zusammengebracht. In der Folge zieht die Donorschicht die Wirkstoffemulsion an und lagert diese ähnlich wie ein Schwamm ein. Der Beladungsvorgang dauert nur wenige Minuten. Er kann ins normale Wasch- und Spülprogramm integriert oder mittels Handwäsche durchgeführt werden. Ausgelöst durch Wärme, Reibung, Feuchtigkeit und Schweiss, setzt der kontinuierliche Entladungsvorgang transdermal ein. Der in der Donorschicht eingelagerte Wirkstoff wird dabei an die Haut abgegeben, wo er seine Wirkung voll entfalten kann. Die Zeitdauer für die Wirkstoffabgabe, die sogenannte Desorptionszeit, ist bei der Herstellung einstellbar und kann über mehrere Stunden anhalten. Die in der Emulsion eingelagerte Wirkstoffmenge wird auf die gewünschte Applikation abgestimmt. Nach dem Tragen werden bei einer erneuten Wäsche allfällig vorhandene Reststoffe vollständig entladen, und das so regenerierte iLoad®-Textil kann mit einer neuen Wirksubstanz wiederbeladen werden.
In dieser Dissertation wurde das oben beschriebene textilbasierte Drug-Delivery-System (DDS) von Grund auf erarbeitet und in die Praxis umgesetzt. Dazu war es notwendig, Hydrogele zu polymerisieren und diese permanent auf Geweben zu verankern. Die Waschpermanenzen der bei den Produktionsversuchen applizierten Donorschicht auf PA, PES, PES/PUE und CO nach 1, 5, 25, 50 und 100 Maschinenwäschen bei 60 °C wurden untersucht und auch nach über 100 Waschgängen für waschpermanent befunden.
Im Rahmen dieser interdisziplinären Arbeit wurde als Nächstes erfolgreich eine Wirkstoffemulsion hergestellt, mit welcher die iLoad®-Textilien problemlos und reproduzierbar mit 1.8 mg Öl / cm2 beladen werden können. Dies entspricht den in der Dermatologie üblicherweise typischen Mengen von auf der Haut applizierten Hautpflegeprodukten. Mittels abschliessend durchgeführten Desorptionsversuchen an humanen In-vitro-Hautmodellen, Reibungsmessungen sowie einem Praxisversuch mit einem Probanden konnte die Wirkstoffabgabe und damit die Funktion des gesamten iLoad®-Systems nachgewiesen werden.
Mit iLoad® ausgerüstete Textilien haben den Zytotoxizitätstest bestanden und gelten somit als hautfreundlich. Alle Komponenten werden dem weltweit strengsten Textilstandard bluesign® entsprechen.

Abstract

Textiles reloadable with active substances for transdermal drug delivery

The iLoad® system, meaning a Reloadable Drug Delivery Textiles (RDDT), consists of fabrics reloadable with cosmetic ingredients or medical drugs for transdermal application of active agents in the field of wellness and health care: iLoad® stands for intelligent loading, release, washing and reloading.
The iLoad® system consists firstly of an eligible base fabric onto which a special donor layer is applied and anchored. In the subsequent loading process, the donor layer (host), i.e. the iLoad® textile, is combined with a specific emulsion (guest) with the individually required, preferably phytotherapeutic active substances. Following that, the donor layer attracts the customized active agent emulsion and stores it like a sponge. The loading process takes just a few minutes and can be integrated into the normal washing and rinsing program or carried out during hand washing. Triggered by warmth, vibration, moisture and perspiration, the constant transdermal unloading process commences and releases the substance in the donor layer onto the skin, where it can develop its full effect. The desorption time, i.e. the length of time for the release of the active agent, can be adapted during production and may continue over hours. The substances stored in the emulsion are tailored to the desired application. After wearing, any residual substances that may be present are completely unloaded in the next wash. Subsequently, the iLoad® textile regenerated in this manner can be loaded with a new active substance.
In this dissertation, the above described textile based Drug Delivery System (DDS) was designed from scratch and realised in practice with the development of hydrogels polymerized and permanently anchored onto the fabrics. To test the washing permanence after 1, 5, 25, 50 and 100 washing machine cycle at 60 °C, the donor layer was applied by production trials to PA, PES, PES/PUE and CO. It has proven stable and permanent even after 100 washing cycles.
Within the scope of this interdisciplinary work, an active agent emulsion was successfully compounded, enabling the loading of 1.8 mg oil / cm2 iLoad® textile without difficulty and in a reproducible way. This correlates with a typically applied amount of skin care product used in dermatology. The performed desorption tests using human in vitro skin models, friction measurements as well as a test in practice with a proband showed the release of the active agent, proving the function of the whole iLoad® system.
Textiles finished with iLoad® easily passed the cytotoxicity test, making them skin-friendly. All the components of the system are subject to the worldwide most stringent textile standard bluesign®.
