UV radiation leads to the formation of cyclobutane pyrimidine dimers (CPDs) and (6-4) photolesions in DNA. These lesions are highly cytotoxic, mutagenic and substantially involved in the development of skin cancer. The interaction of these lesions with the most important DNA-dependent processes in the cell, such as replication, transcription and DNA repair, has so far been investigated at the atomic level in only a few cases. This work comprises mechanistic studies on the reductive repair of these photolesions and their analogues in DNA strands, which are of fundamental importance for understanding reductive electron transfer in DNA. A further part of the work deals with studies on the recognition of the most frequent photolesion, the cyclobutane pyrimidine dimer (CPD), by enzymes such as CPD photolyase or eukaryotic RNA polymerase II. In addition, work was carried out on the synthesis of the (6-4) photolesion and a 15N-labelled derivative in order to investigate the repair mechanism of (6-4) photolyase.
