Fluorescence spectroscopic techniques have not only become established as standard methods of analytical chemistry; they also open up profound insights into biochemical processes in vitro and in vivo. A pressing task of method-oriented research is the development of new analytical procedures tailored to specific applications that take account of the enhanced capabilities of modern, multidimensional fluorescence spectroscopy. This work presents not only the further development of such techniques and evaluation routines, but also demonstrates the performance of these methods in several fields of application.
One focus is the investigation of protein structure, dynamics and aggregation. Thus, a new analysis strategy for time- and emission-wavelength-resolved laser-induced fluorescence (LIF) is presented, by means of which relaxation processes in single-tryptophan proteins can be detected in the nanosecond range. In fluorescence microscopy, hyperspectral fluorescence lifetime imaging (hs-FLIM) enables the separation of the fluorescence contributions of spectrally very similar fluorescent labels. In addition, the Parkinson-causing aggregation of a-synuclein is observed in vitro and in human neuronal cells.
Multidimensional optical spectroscopy is ideally suited to characterising the photophysical properties of novel compounds. This is demonstrated by way of example in the investigation of luminescent benzodiazaboroles, which exhibit high quantum yields in the blue spectral region and are thus attractive emitters for use in organic LEDs. By combination with more Lewis-acidic triarylborane units, fluoride-sensitive fluorescence switches can be generated.
