Kai Lieder’s dissertation “Interface-Peptide der viralen Protease von HIV-1 als gentherapeutisch einsetzbare antivirale Substanzen” investigates novel approaches to inhibiting HIV-1 replication through intracellularly expressed peptides. The research focuses on antiviral compounds designed to interfere with the dimerization interface of HIV-1 protease rather than its catalytic active site. Various interface peptides are expressed in eukaryotic cell lines using genetically engineered expression vectors. Their antiviral activity is evaluated through viral production, infectivity assays, and analysis of viral precursor protein processing. The findings demonstrate that certain peptides inhibit HIV-1 replication, although different inhibitory mechanisms appear to be involved. A second research objective concerns the targeted delivery of intracellularly expressed proteins into newly forming viral particles. Using viral targeting sequences and EGFP fusion proteins, the study demonstrates the incorporation of nonviral proteins into HIV-1 particles. These findings provide an experimental foundation for developing gene therapeutic strategies and alternative antiviral approaches targeting HIV-1 replication.
