The aim of this dissertation was the in vitro reconstruction of various vascular barriers. The microfluidic μ3DVasc bioreactor was used as an in vitro model for a vascularised, three-dimensional (3D) tissue. This bioreactor, based on SMART technology, consists of a porous microcapillary for culturing vascular endothelium and an underlying chamber surrounding the capillary for a 3D tissue. Three barriers were selected: the blood–brain barrier, the intestinal epithelium and the liver. All models were established in 3D and validated in comparison with the 2D models and the natural in vivo barriers.
In future, the 3D models presented in this work in the μ3DVasc bioreactor offer a suitable platform for investigating drug uptake as well as the distribution and accumulation of the applied substances in tissues. Linking the three models to one another would create numerous innovative investigative possibilities, such as testing newly developed substances efficiently and cost-effectively in an HTS format in order to identify suitable candidates more rapidly, optimise them further and ultimately bring them to market.
